Effect of folinate on thymidine uptake by Pediococcus cerevisiae.

Uptake of (3)H-thymidine by resting cells of Pediococcus cerevisiae was found to be energy- and temperature-dependent. The pH optimum was between 6.5 and 8.0, and after 2 min of incubation most of the radioactivity was found in the deoxyribonucleic acid (DNA) fraction. Iodoacetate at a concentration of 10(-2)m caused a 50% inhibition of uptake. Preincubation of resting cells for 10 min with fol...

Transport of folinate and related compounds in Pediococcus cerevisiae.

The properties of folinate and 5-methyltetrahydrofolate (5-CH(3)-H(4)PteGlu) transport mechanism of Pediococcus cerevisiae were studied. The uptake was dependent on temperature, pH (optimum for both compounds at pH 6.0), and glucose. Iodoacetate, potassium fluoride, and sodium azide inhibited the uptake. 5-CH(3)-H(4)-PteGlu was apparently not metabolized but folinate was metabolized. Metabolism...

Effects of uracil and thymidine on the development of resistance to 5-fluorouracil in Pediococcus cerevisiae.

Bacteriol. 85:97-105. 1963.-Growth of Pediococcus cerevisiae ATCC 8081 in a basal semidefined medium was inhibited by 5-fluorouracil (FU) and 5-fluoro-2'-deoxyuridine (FUDR). Addition of uracil slightly d...

Uptake and Metabolism of Folate by a Folate-utilizing Mutant of Pediococcus cerevisiae

Pediococcus cerevisiae 8081, which uses folinic acid for growth, and its nutritional mutant 8081/S, which utilizes folate, were compared with respect to their ability to take up and metabolize folate. In contrast to the parent strain 8081, which takes up very little folate, the uptake of folate by the mutant is a concentrative, saturable, and energydependent process. At low extracellular concen...

Physiological and enzymatic properties of a thymidine-requiring Pediococcus cerevisiae mutant.

We describe the isolation and characterization of a Pediococcus cerevisiae thymidine-requiring mutant and its thymidine-independent revertant. The mutant strain lacked thymidylate synthetase activity and had an absolute requirement for low concentrations (2 micrograms/ml) of thymidine in addition to a requirement for N-5-formyl tetrahydrofolic acid (folinate). Even at high concentrations (up to...
